売れない若手漫才コンビが巻き起こすハチャメチャな大騒動! 関西ジャニーズJr.第4弾映画! 涙と笑いの"なんでやねん!"な青春コメディ。売れない漫才コンビ「エンドレス」と漫才トリオ「ピンクらくだ」がライバルとして競い合い、悩みながら、互いに夢に向かって成長していく姿を、ボケあり、ツッコミあり、涙ありで描く。物語の中心となる漫才コンビ「エンドレス」を演じるのは、前作でも主演を務めた西畑大吾と藤原丈一郎。2人のライバルとなる「ピンクらくだ」のメンバーを、向井康二、室龍太、草間リチャード敬太という個性を持った3人が熱演する。更に、舞台で活躍中の浜中文一のほか、正門良規、小島健、道枝駿佑、長尾謙杜、高橋恭平、嶋崎斗亜ら若手メンバーがフレッシュな魅力を放つ。――高浜優輔と稲毛潤は幼馴染み。人を笑わせるのが大好きなふたりの夢は、「お笑いでみんなを幸せにする」こと。夢を叶えるため、お笑い養成所に通い始めたふたりは「エンドレス」というコンビを組み、「ピンクらくだ」とライバルとして競い合っていた。遂に、若手芸人の登竜門コンテストで優勝したエンドレスは、TV局からも声が掛かり始め人気が爆発!! しかし、「売れることが第一」と考える高浜と、純粋にお笑いが好きな稲毛との間が次第にギクシャクし始める。そんな折、ライバルのピンクらくだが突然ブレイク。エンドレスは人気も下降し、高浜はイラ立ちを隠せない。ある日のこと、高浜は稲毛にお笑い強化合宿をしようと提案する。ところが、その場所は幽霊が出るとウワサの旅館。なぜかピンクらくだも怪しい霊媒師と心霊スポットロケで同じ旅館に泊まることになり・・・。
