ハングオーバー! の脚本コンビが贈る、ハリウッド最新入れ替わりコメディ! ――デイヴ・ロックウッド(ジェイソン・ベイトマン)は、弁護士として一流法律事務所で働き、美人の妻ジェイミー(レスリー・マン)や、3人の子供とともに、瀟洒な邸宅で暮らす男性だ。良き家庭人である彼は、理想の人生を手に入れたように見える。とは言え、本人にしてみれば、家庭と仕事への責任に縛られた人生には、やや疲れを感じているご様子。デイヴの幼馴染で親友の、ミッチ・プランコ(ライアン・レイノルズ)は、また、正反対の人生を送っている。ミッチは未だ独身で、俳優になるという夢に生きているようだ、それでも、時折、CMなどの小さな仕事のオーディションを受ける程度のもの。ただ、ハンサムな独り身である彼の周囲には、かならず女性が寄り添っていて、そちらの生活はなかなか熱いようである。そんな2人が、ある日、とあるスポーツバーで酒を酌み交わし・・・。
