ラスト5分、かつてない極限状態にあなたは耐えられるか? ジェームズ・キャメロン製作総指揮、究極の体感サバイバル・アドベンチャー! ――パプア・ニューギニアの熱帯雨林の奥地に存在する、巨大な洞窟体系[エスペトリ・エサーラ]。地球上で最も大きく、最も美しく、最も近づきがたいその場所は、人が足を踏み入れてはいけない聖域(サンクタム)として君臨していた。そんな洞窟の謎を解き明かそうと、大富豪の実業家カールのスポンサードを受け、経験と判断力を誇る冒険家フランク(リチャード・クロスバーグ)率いる探検隊が、ハイテク装備とダイビングを駆使した調査に挑む。しかし複雑に入り組んだトンネルがどこへ通じているのかをいっこうに解明できず、メンバーは苛立ちを募らせていた。とりわけ、フランクによって強引に探検に参加させられた息子のジョシュは、重労働を命じられうんざりする毎日。そんなある日、突然襲来した巨大なサイクロンの影響で、すさまじい鉄砲水が洞窟を襲い、メンバーは唯一の出口を塞がれてしまう。生き延びるため、暗く冷たい洞窟の奥へ奥へと進むことを余儀なくされた探検隊の行く手を待ち構えていたのは、美しくも恐ろしい自然の驚異。迫りくる激流と戦い、迷路のようなトンネルを這い、断崖絶壁を降下し、闇に包まれた水中に潜っていくしかなかった。果たして、メンバーの無謀ともいえる試みの先に待ち受けているものとは?
