本作はAmazonプライム・ビデオで2016年4月1日から全13話で配信された〈Season1〉、2017年4月7日から全13話で配信された〈Season2〉をそれぞれ劇場公開用に編集、再構成。「覚醒」「輪廻」とテーマ性を重視したストーリー構築を施し、新たな映画として公開。「生と死」を物語の中で重要なテーマとして扱い、「養殖と野生」をコンセプトに、病弱でひきこもりがちな青年、水澤悠(はるか)(藤田富)/仮面ライダーアマゾンオメガと、アマゾンを狩るアマゾンとして獣のように生きる男、鷹山仁(谷口賢志)/仮面ライダーアマゾンアルファを中心に物語が展開。その過激な描写やハードなアクションは、仮面ライダー作品の中で異色と評された。
